Dear Pailin Nieuwenhuis,

Although the methods to determine water absorbing capacity have not
clearly
mentioned in original note
(http://www.gramene.org/newsletters/rice_genetics/rgn4/v4p95.html), it
should be
done like as follows:
1. Putting certain amount of culture solution or water in the container
(pot);
2. Planting mutant plant and wild-type plant in separate containers, and
placing
containers under normal growth condition, usually in greenhouse;
3. After 6 hours and 16 hours, determining culture solution amount left
in the
containers and calculate absorbed amount by plant; (For total roots this
time)
4. Cutting short roots and only keeping longest root for both mutant and
wild-type
plants, repeating above procedure (1 to 3), you can get water uptake
amount for
longest root.
